Chichester was the son of Sir Robert Chichester, K.B., of Raleigh, Devon and his second wife Mary Hill, daughter of Robert Hill of Shilston, Devon. He succeeded to Raleigh on the death of his father on 24 April 1624. He was created a baronet on 4 August 1641.〔(George Edward Cokayne ''Complete Baronetage, Volume 2'' )〕 In 1661, Chichester was elected Member of Parliament for Barnstaple in the Cavalier Parliament and sat until his death in 1667 at the age of about 44.〔(History of Parliament Online - Chichester, Sir John, 1st Bt. )〕 Chichester married firstly, Elizabeth Rayney, daughter of Sir John Rayney, Ist Baronet and his first wife Catharine Style, daughter of Thomas Style. She died in 1654 and he married secondly Mary Warcup, widow of St. Anne's, Blackfriars, at that parish on 18 July 1655.
